As always your vectors are of excellent quality and replicate the outlook from the show flawlessly. Your quality is undeniable. Technique: 5 stars.
Taking into account that the joke is a twist of an existing scene and that the idea was proposed by someone else that hurts the Originality score a bit. Something that is not always the case in your work, that, while kinky, I have always found it new and original, pushing boundaries of Pony Vector Art. Originality 4 stars.
Your goal in this piece was clear, and was conveyed without fault. Yes it was a joke, but the delivery was very good. Focusing on Twilight with her flank exposed in the two last frames allowed the punch to develop while setting the undertone of the joke. Vision 5 stars.
The idea behind the joke was nice, and got a chuckle out of me. Twilight's expression in the last panel is funny.
However ,while I can't think of a better one, it seems to me that Molestia's line was a little mild.
And the horrific realization in Twilight's face could be more extreme. Perhaps you could use smaller irises and bigger eyes?
As it is, while funny, it lacks some "punch". Unless you were aiming for subtlety.
4 stars for impact.
